After his reinstated 800m gold on Sunday, Weir saw off Thai Prawat Wahoram and Australia's Kurt Fearnley in treacherous wet conditions.
Shelly Woods won 1500m silver, while Mickey Bushell was 0.09 seconds behind American Josh George in the T53 100m.
Tuesday's efforts on the track meant ParalympicsGB had won over 100 medals.
A late burst pushed Woods into second in the 1500m T54, as Canadian Chantal Petitclerc, 38, claimed her fifth gold.
Hazel Simpson's bronze in the T36 100m was her second Games medal, with David Weir and Oscar Pistorius racing later.
Woods, bronze medallist from the re-run 5000m, said: "I had some speed at the end. I'm so happy."
Canada's Chantal, picking up her fifth title in Beijing and her 13th career Paralympic gold, said: "Athens was great and this was more than great.
"Everything was more difficult - the girls are fitter and I'm getting older."
Earlier on Tuesday, Britain's Nathan Stephens agonisingly missed out on a F57/58 javelin bronze.
The 20-year-old was in third before Czech thrower Rostislav Pohlmann surpassed the Cardiff student's 38.56m.
"I'm disappointed because I was so close to the bronze but this is a mixed category event and I'm an F57 so to come fourth is pretty awesome," he said.
"It was daunting for a little 20-year-old against the big guys. I'm still a baby compared to them. This is my first Paralympics and my main aim has always been London so to come fourth is fantastic."
GB's Danny Nobbs was 10th in the F53/54 shot put with a best throw of 9.13m.
Libby Clegg qualified for the 200m B final after finishing fourth in her T12 semi in 26.16s.
South African sprint double winner Pistorius is looking for his third gold of the Games in the 400m T44 final, with Ian Jones also in action.
